What Is Spravato?
Spravato is a prescription nasal spray containing esketamine. It is FDA-approved for adults with treatment-resistant depression (TRD)—used on its own or with an oral antidepressant—and, together with an oral antidepressant, for depressive symptoms in adults with major depressive disorder who have acute suicidal thoughts or actions.

Who May Be a Candidate?
You may be a candidate if you:
- Are 18 or older with major depressive disorder
- Have tried at least two antidepressants without adequate improvement
- Can attend in-clinic sessions and arrange a ride home
Spravato is not appropriate for people with aneurysmal vascular disease, arteriovenous malformations, a history of bleeding in the brain, or an allergy to esketamine or ketamine. Your provider will also review blood pressure, heart and lung health, pregnancy, and any history of substance use or psychosis.
How Spravato Works
Most antidepressants target serotonin. Esketamine acts on glutamate, the brain's most abundant chemical messenger, by blocking NMDA receptors. Researchers believe this helps restore connections between brain cells involved in mood. Some patients notice changes within days to weeks, though everyone responds differently.
What a Session Looks Like in Ripley
